How to use the meganuclease platform
The goal is to induce efficient DNA targeted recombination at a precise locus in living cells. Clients provide Cellectis with a piece of the DNA sequence (between 10 to 50 kb) or complete genome to be targeted and position of the preferential target site. Cellectis processes the sequence and engineers a custom Meganuclease Recombination System (c-MRS) to be used by the client to induce the desired genetic changes in the preferred species. Cellectis provides technical support and Genome Engineering consulting during the entire process.
Cellectis Omegabase is a comprehensive database of meganucleases that have been tested against a series of DNA targets. Our high-throughput meganuclease platform, and the modularity of the meganucleases facilitate the identification, generation and engineering of Custom Made Meganuclease for inducing the desired genetic changes in the preferred species
Either Omegabase search leads directly to a validated Meganuclease readily available for this DNA target. Alternatively, the search produces potential Meganucleases that need to be validated and/or re-engineered before delivery to our client.
As of today, Cellectis offer related to c-MRS includes the following scenarii:
I- Engineer for a Client a c-MRS to a DNA target of value: A large sequence file with a preferred hit point is sent to Cellectis. Cellectis proposes a series of reachable targets around the hit point, if any. After decision on privileged target(s) to start with, c-MRS is validated in CHO cells before testing in designated organism.
II- Select a target from current Cellectis Omegabase: Client browses through Cellectis Omegabase. Client selects potentially reachable DNA target(s). Cellectis retrieves the set of meganucleases and engineers a c-MRS. Candidate c-MRS validated in CHO cells to be tested in designated organism.
III- Challenge Client proprietary DNA sequence database to the Omegabase: Sign up for the Omegabase current DNA target hit potential. The Omegabase is searched for Client database sequences. Client seeks potentially reachable DNA targets: If DNA target is selected we proceed as in # II. If not the evolving Omegabase is rechallenged on a quarterly basis.
