How to use the meganuclease platform

The objective is to induce an effective DNA recombination at a specific genomic locus of a living cell. Cellectis analyses the target DNA sequence (this might go as far as the analysis of an entire genome) in which the modification is to take place, and select the preferred localization of the target sites. Target sequences enable the development of a meganucleases recombination system, which can be used to induce the genetic modifications required in a given species.

Cellectis has developed a comprehensive database known as the “Omegabase”, which contains meganucleases that have already been tested with target DNA sequences. This high-throughput meganuclease screening platform, along with the modularity of the meganucleases, facilitates the identification, generation and engineering of meganucleases that can induce the genetic modifications required in a particular species.

A simple search in the Omegabase can identify a meganuclease that has been validated and is ready for use for a given DNA target sequence. A search in the database can also point to potential meganucleases that are yet to be validated and/or whose design needs to be reviewed before validation.

The Omegabase is a database of new meganucleases generated by Cellectis. It is in constant development, both in terms of size and performance.
